Survey of the didemnins: A class of depsipeptide natural products with promising biomedical applications
نویسنده
چکیده
The didemnins represent a versatile class of depsipeptides of marine origin and hold a great deal of potential for biomedical application. The biological and geographical origins of the didemnins are reviewed in addition to the chemical structures of the major didemnins. The biological mechanisms behind the antiviral and anticancer effects of selected didemnins are summarized and the special case of dehydrodidemnin B (Aplidin ) is expounded upon including structural characteristics, synthesis, pharmacological mechanism and a discussion of its current clinical trials as an anticancer agent.

Structural basis for the binding of didemnins to human elongation factor eEF1A and rationale for the potent antitumor activity of these marine natural products.
Didemnins and tamandarins are closely related marine natural products with potent inhibitory effects on protein synthesis and cell viability. On the basis of available biochemical and structural evidence and results from molecular dynamics simulations, a model is proposed that accounts for the strong and selective binding of these compounds to human elongation factor eEF1A in the presence of GT...
متن کاملPersian Gulf Bivalves: Bioactive Pharmaceutical Compounds and Biomedical Applications
Bivalves are a class of marine mollusks with high nutritional value in addition to various medicinal products. The Persian Gulf is home to 224 species from 29 families of bivalves identified so far. Based on research conducted in the Persian Gulf or other parts of the world, bioactive compounds exist in their shell and soft tissue. In this review article, we reviewed biomedical research related...
متن کاملThe first biologically active synthetic analogues of FK228, the depsipeptide histone deacetylase inhibitor.
The FK228 and spiruchostatin bicyclic depsipeptide natural products are among the most potent histone deacetylase (HDAC) inhibitors known. Although FK228 is in advanced clinical trials, the complexity of the natural products has precluded mechanistic studies and the discovery of structure-activity relationships. By total synthesis, we have prepared the first depsipeptide analogues. Our results ...
متن کاملA Review of Aerogel Applications in Adsorption and Catalysis
Aerogels are a special class of porous material, which have excellent physicochemical properties such as low density, high porosity, high surface area and adjustable surface chemistry. Aerogels were first prepared several decades ago, but never truly commercialized due to their high cost. Technological advancements in the production and quality of different types of aerogel cut costs down and m...
متن کامل1 An Update on the Biomedical Prospects of Marine-derived Small Molecules with Fascinating Atom and Stereochemical Diversity
In this chapter we discuss a selection of structurally diverse marine-derived small molecules (MDSMs) with potent and/or specific bioactivity and analyze their biomedical applications. The compounds included have been isolated either from marine macroorganisms, including sponges, ascidians (tunicates), bryozoans, and molluscs, or from microorganisms, such as bacteria and fungi. Our inquiry begi...
